SMTP routing over VPN   
We recently had two separate locations with two separate Exchange
servers on two separate domains: site1.local and site2.local

We just installed a VPN, and now each location can ping the other and
access server shares. (We did not change domains or trusts or anything
on either end.)

Our primary (public) Exchange server is on site1, so all users on
site1's Exchange server have .com email addresses which work great.

But if a user from site2 sends an email to someone on site1, and the
site1 recipient attempts to reply, we get a "No route was found for
the recipient server." response.

  To:      test@site2.local
  Subject: Test
  Sent:    Mon, 30 Jun 2008 14:14:10 -0500

did not reach the following recipient(s):

test@site2.local on Mon, 30 Jun 2008 14:14:15 -0500
    No route was found for the recipient server.  Please contact your
system administrator.

How can this be corrected, please.

Can you telnet to port 25 of the server in site1 from site2?
